Study on Rapid Propagation Technology of Tissue Culture of Dioscorea alata ‘Minghuai No.1’

  • Purpose The aim was to establish rapid propagation of Dioscorea alata ‘Minghuai No.1’.Method With the tender stems of Dioscorea alata ‘Minghuai No.1’ as explants, we carried out different hormone proportioning optimization research of ‘Minghuai No.1’ primary culture, proliferation and rooting culture,by uniform design and regression analysis method.Results The optimal axillary buds induction medium was MS+6-BA 2.0 mg/L+NAA 0.1 mg/L+PVP 100 mg/L+agar 4.0 g/L+sugar 30 g/L, the induction time was short and the induction rate was 92%, which could induce the formation of cluster buds; the optimal subculture medium was MS+6-BA 2.33 mg/L+NAA 0.1 mg/L+PP333 0.2 mg/L+agar 4.0 g/L+sugar 30 g/L, and the proliferation coefficient could up to 3 times; The optimal rooting medium was 1/2 MS+IBA 0.34 mg/L+IAA 0.5 mg/L+AC 1.0 g/L+agar 4.5 g/L+sugar 20 g/L, the rooting rate was 95%, and the rooting effect was good.Conclusion The experimental results laid a good foundation for the promotion and industrial development of ‘Minghuai No.1’.
