Molecular Identification of the Root-knot Nematodes Species in the Parts of Southwest China

  • In this study, we detected and identified 19 root-knot nematodes populations on the crops from part areas of Yunnan and Guangxi Provinces using rDNA-ITS region and SCAR primers. According to the analysis of the tested samples, it indicates that the length of ITS sequence was about 750 bp and the sequence homologies were 96.25%. The gene distances of tomato from Yuanmou (RKN-10) and tomato from Honghe college (RKN-13) to Meloidogyne enterolobii were very close with high sequence homologies. Moreover, the gene distances of Meloidogyne incognita, Meloidogyne javanica and Meloidogyne arenaria to the other 17 root-knot nematodes populations were very close with high sequence homologies. It is identified that there were three M.javanica, six M.incognita and eight M.arenaria among 17 root-knot nematodes populations by SCAR primers. The phylogenetic tree of ITS region showed that RKN-10, RKN-13 and Meloidogyne enterolobii get together. There was a close genetic relationship between them and the rest of the root-knot nematodes populations get together. The genetic relationship between Meloidogyne incognita and Meloidogyne arenaria were more closer than Meloidogyne javanica.
