Study on Tumor Inhibition of Dichloromethane Extract from Huang Yao (Blumea balsamifera DC.) of Yi Nationality in Lewis Lung Cancer Mice

  • Huang Yao (Blumea balsamifera DC.) of Yi nationality is widely used for treatment of lung cancer in minority areas,and the early research found that the dichloromethane extract from it has better anti-tumor activity on A549 cells.Therefore,on the basis of preliminary studies,we tried to study the anti-tumoreffect of the dichloromethane extract from Huang Yao of Yi nationality on Lewis lung cancer in mice.Through the establishment of model of C57BL/6J mouse with Lewis lung cancer,we observed the inhibitory effect of the dichloromethane extract from Huang Yao; and used H.E. staining method to study the pathological changes in the tumor tissue; and applied the immunohistochemistry (IHC-P) method to detect the expression of VEGF and TNF- protein in tumor tissues.The results showed that compared with the model group, the dichloromethane extract from Huang Yao could significantly inhibit the growth of Lewis lung carcinoma in mice (P 0.05 or P 0.01). The result of H.E.staining showed that the tumor tissue necrosis was obvious, the cell density was decreased, and the nuclear condensation and fragmentation was severe in the treatment group compared with the model group, which high dose group is the most obvious; the positive expression of VEGF and TNF- was detected by IHC-P in the cytoplasm of tumor cells, and the average optical density of VEGF and TNF- in each group was significantly higher than that in the model group (P0.05). The experiment showed that the inhibition in mice with Lewis lung cancer was significant of the dichloromethane extracts from Huang Yao of Yi nationality, and the anti-tumor effect may be associated with the down-regulation of the expression of VEGF and TNF- protein.
