Study on Genetic Diversity for Yunnan Popular Maize Inbred Lines by SSR

Abstract
To define genetic relationships for maize inbred lines is obviously important to germplasm utilization and new hybrid development. Improved CTAB DNA extraction method, SSR molecular marker, and native gel electrophoresis silver staining technique were used in the experiment; data were analyzed by Excel 2007 and clustered by NTSYS-pc 2.10e software,respectively. Seventy SSR primers were screened to analyze genetic diversity for 19 Yunnan popular inbred lines, the results showed that totally 504 polymorphic amplified fragments were detected, and each primer pair amplified 2 to 13 variant alleles with an average of 7.21. The polymorphism information content (PIC) for per primer varied from 0.40 to 0.90 with an average of 0.73. Genetic similarities among 19 inbred lines were 0.611 to 0.841 with an average of 0.703. And 19 inbred lines were divided into 4 groups according to their genetic similarities by bounded coefficient of 0.697 with UPGMA method, the results indicated that Yunnan maize inbreds had rich genetic diversity and polymorphism; it provided references for Yunnan maize germplasm improvement, heterotic grouping and hybrid development.
