Classification of Indica-Japonica and Optimization of Embryonic Callus Inducing Medium in a Perennial Rice (Oryza sativa L.)

Abstract
There is high research value in perennial rice on account of it's excellent biological characteristics, whereas, people commonly use japonica rice as genetic receptor in genetic engineering for improvement of rice. In this study, we identified indica-japonica classification of a perennial rice (Oryza sativa L.) by means of InDel molecular marker initially, then we conducted an orthogonal test which estimated by inducing rates and qualities of its embryonic callus to optimize the culture medium, in order to provide a technical support for its further genetic improvement and relative gene functional research. Our experimental results indicated that PHR-1 is a typical japonica rice, for which the optimal embryonic callus inducing medium is consisted of 30 g/L sucrose+N6 macroelements +N6 microelements+N6 organics+molysite+0.5 g/L casamino acid+2 g/L L-proline+3 mg/L 2,4-D.
