Application of Western-blotting Technique to the Study of Low Temperature Stress on Antioxidant Enzyme Activity of Cassava Leaves

Abstract
In order to study the effect of low temperature stress on expression levels of antioxidant enzymes in cassava leaves, cassava cultivar South China 205(SC205) were used as the materials, expression level of ascorbate peroxidase (APX), glutathione reductase (GR), superoxide dismutase (SOD) and peroxidase (POD) in leaves during 7 d after exposed to 5℃ were measured by Western-blotting technique. The results showed that:in the early phase, the expression levels of 4 kinds of enzymes were increased with stress time, SOD and GR expression was the highest at stressed for 3 d, while APX was at 5 d and POD was 4 d. After reaching the highest, APX and SOD expression levels were fall after rising, while GR and POD was declined. The results and method can provide a reference for further understanding of the physiological mechanisms on low temperature tolerant in cassava.
