Effects of Four Different Media on Spore Germination and Gametophyte Development of Sphaeropteris brunoniana

Abstract
Spore germination and gametophyte development of Sphaeropteris brunoniana under four different media were observed with an optical microscope. Spores cultured on medium MS and 1/2 MS couldn't germinate; spores cultured on medium 1/4 MS germinated within 285 days after sowed; while spores cultured on 1/8 MS germinated within 40 days after sowed. There were also differences on many aspects such as the appearance time and the rates of filaments, platelets, cordate prothallia, antheridia and archegonia under different media. Spores cultured on 1/8 MS germinated and developed about seven times earlier than that cultured on 1/4 MS, and the ratio was more than that. The results showed that the high salt medium was not suitable for spore germination and gametophyte development of S.brunoniana.
