Abstract
In this study, a multiplex PCR detection system was established for Xanthomonas oryzae pv. oryzicola and Xanthomonas oryzae pv. oryzae by designing two specific primers. Through system optimization, specificity and sensitivity tests, the results showed that two pairs of specific primers had a high specificity and sensitivity, and the detection sensitivity of X.oryzae pv. oryzicola and X.oryzae pv. oryzae were 104 cfu/mL and 105 cfu/mL,respectively. Using the multiple PCR system established in this study, a rice variety seed with X. oryzae pv. oryzicola and another rice variety seed with X. oryzae pv. oryzae were successfully detected from 50 kinds of commercially available rice seeds. A multiplex PCR detection system established in this study provides the effective detection technology and means for the quarantine department to control the invasion of these two kinds of quarantine seedborne diseases.
