Analysis of the Impact of Tobacco Structure on Puff-by-puff Release of NNK in Mainstream Smoke

Abstract
In order to study the relationship between NNK release puff by puff in mainstream cigarette smoke and tobacco structure, we designed and made samples of different types of tobacco and cut tobacco composition. Through the by mouth suction unit linked with RM20H smoking machine, we captured smoke and particulate matter automatically in mainstream smoke, and with reference to national standards, we used HPLC-MS/MS to realize the quantitative analysis of NNK release and trends. The results show that (1) The proportion of tobacco structure had a greater impact on overall NNK release of cigarette, adjusting formula and reducing burley tobacco could effectively reduce the proportion of total releases NNK. (2) NNK's release by mouth with a suction port increased linearly with sequence trend, but the release rapidly declined after the 5th mouth; In the previous 5 pumping process, a high proportion of burley tobacco sample or proportion of normal cut tobacco had a higher release by mouth; At 6th mouth, the influence from proportion of burley and normal cut tobacco gradually declined. (3) There was a strong linear regression relationship among types of tobacco, cut tobacco composition and NNK puff-by-puff release (1~6), the correlation coefficient (R) was range from 0.951 to 0.999, by appropriate reducing the proportion of burley, increasing the proportion of expanded tobacco could reduce the release of NNK puff-by-puff of the previous 5 mouths.
