Analysis on Genetic Diversity of Apis cerana in Yunnan Province Using Mitochondrial DNA COⅠ~COⅡ

  • To analyze the genetic diversity of Apis cerana in Yunnan Province, the region between COⅠ~CO Ⅱ of the mitochondrial DNA (mtDNA) was chosen as an index, 179 populations in 6 sampling sites were sampled and analyzed by bioinformatics softwares. The results revealed that: (1) the total length of these mtDNA fragment was 760 bp, the base composition of A, T, C and G in the sequences was 40.4%, 42.4%, 10.7% and 6.5%, respectively, which suggested a strong A+T bias.(2) There were 28 variable sites,18 of them are parsim information sites,the others are singleton sites. (3) A total of 29 haplotypes were identified, the haplotype diversity was 0.884, and the nucleotide diversity was 0.002 69.(4)The results of cluster analysis showed that the A.cerana of Yunnan Province had no obvious differentiation and the genetic relationship of A.cerana was closer to those A.cerana samples from Chongqing,Gansu,Beijing than those samples from Taiwan Province,while the genetic relationship was closer to those A.cerana samples from Japan and South Korea than those samples from Malaysia .
