Molecular Evolution Analysis of Cell Division Related Gene Ndc80

  • The majority of tumor cells show chromosome instability. In the process of mitosis, whether kinetochore-microtubules are right combination has important relationship with the stability of the chromosome. Ndc80 complex is composed with Ndc80, Nuf2, Spc24 and Spc25 which play key roles in kinetochore-microtubules combination. In this study, the molecular evolution of Ndc80 a total of 16 kinds of biological gene represent fungi, invertebrates and vertebrates were analyzed by means of using biological information. The result showed that, open reading frame changes of Ndc80 gene from the fungus to vertebrates are relatively small, most organisms have a tendency to TGA as termination codon, Ndc80 gene exon from less to more of the process, and finally tends to have 17 exons. Amino acid sequences in mammalian are highly homologous, full lengths are more conservative. The above results suggest that Ndc80 is a more conservative gene, plays an important role in kinetochore-microtubule connection and its stable expression plays a key role in stability of chromosome and tumor suppressor.
