Preliminary Identification of Horticultural Plant Spotted Wilt Diseases in Kunming

  • Tospovirus is the only genus to infect plants in the Bunyaviridae family. During 20122013, 13 samples that possibly infected by Tospovirus were collected from scenic landscaping and vegetable-growing field in Kunming, Yunnan Province. The complete nucleic acid sequence of nucleocapsid protein (NP) was obtained by using RT-PCR, cloning and sequencing.The phylogenetic tree was constructed using MEGA 5.05 software based on the amino sequence of NP. The result showed that 11 of 13 samples were identified as Tospovirus, including Tomato spotted wilt virus(TSWV), Calla lily chlorotic spot virus (CCSV), Tomato zonate spot virus(TZSV), Impatiens necrotic spot virus (INSV) and Hippeastrum chlorotic ringspot virus (HCRV). The natural hosts are Hymenocallis littoralis, Phalaenopsis amabilis, Lycopersicon esculentum, Alocasia culcullata, Datura stramoniumL.,Tropaeolum majus,Iris tectorumand Philodendron andreanum. A. culcullata is a new host of compound infection of Tospovirus.
