Effects of Natural Antioxidant Extracts on the Molecular Mechanism of Anti-aging in HDF

Abstract
The purpose of this study is to investigate the effects of natural antioxidants (grape seed extract, green tea extract, green tea polyphenols, Pu-erh tea extract, Pu-erh tea polysaccharide) on the molecular mechanism of anti-aging in old cell model of HDF. After establishment of old cell model, the HDF cells were cultured in the medium containing different concentrations natural antioxidants with low concentration (3.125, 6.250 g/mL), medium concentration (12.500, 25.000 g/mL) and high concentration (50.000, 100.000 g/mL) from Day 1 to Days 7. The ability of HDF proliferation was determined by using MTT method. Total RNA was extracted from each culture treatment to determine expressions of SOD1, SOD2, CAT, GPx, mitochondrial D310 and D-loop genes. It has been shown that the low concentration treatments of all natural antioxidants significantly increased the proliferation ability in old HDF. In medium and high concentration treatments, only the treatments from Pu-erh tea polysaccharide and Vc were observed to increase significantly proliferation ability of HDF. The green tea extract and green tea polyphenols treatments significantly up-regulated expressions of SOD1, SOD2, mitochondrial D310 and D-loop genes of old HDF cell. Pu-erh tea treatments significantly up-regulated expressions of SOD1, GPx and D-loop genes while Vc treatments significantly up-regulated expressions of SOD1 and GPx genes old HDF cell. All natural antioxidants treatments from current study were observed to have anti-aging function. Compared with others, Pu-erh tea polysaccharide showed high ability of anti-aging in HDF cell. It has been shown that there were different molecular mechanisms or pathways for the natural antioxidants affecting on anti-aging functions in HFD cell.
