Abstract
In this study, the gene of Gastrodia antifungal protein(GAFP) was cloned and transferred into one cultivar of land cotton using the method of pollen-tube-channel, with the purpose of obtaining transgenic plants with improved resistance to wilt. Five transgenic cottons, including 672GAFP-1, 672GAFP-2, 672GAFP-3, 672GAFP-4 and 672GAFP-5, were obtained and proved based on RCR and RT-PCR. After breeding for two years, the progeny of 672GAFP-1, 672GAFP-2 and 672GAFP-5 lines showed a stable and strong resistance to Verticillium wilt and Fusarium wilt. Field analysis showed that the transgenic cotton yield increased by 30% more than that of non-transgenic, agronomic characters and fiber quality analysis showed that the transgenic cotton was better than non-transgenic in disease nursery, but they were not different in field of free disease. Lines with high levels of resistance to Verticillium wilt and Fusarium wilt obtained from the present study may be widely planted and help to reduce the future impact of cotton wilt on production resulting in increased yields.
