Regulatory Effects of Moringa oleifera Seeds Water Extract on Jejunal Mucositis Induced by 5-Fluorouracil in Mice

  • Purpose To explore the improvement of Moringa oleiferia seeds water extract on jejunal mucositis induced by 5-fluorouracil (5-FU) in mice.
    Methods A total of 50 SPF grade male Kunming mice were randomly divided into normal group, model group (intraperitoneally injected with 130 mg/kg 5-FU) and low dose, medium dose and high dose of M. oleiferia seeds water extract groups (administrated with M. oleiferia seeds water extract 50, 100 and 200 mg/kg, respectively). On the 1st day to the 14th day of the experiment, normal group and model group were given distilled water, M. oleiferia seeds group was given different doses of M. oleiferia seed water extract every day; on the 15th day to the 16th day, normal group was intraperitoneally injected with normal saline, and other groups were intraperitoneally injected with 5-FU every day; observation was carried out on the 17th day to the 18th day; the mice were killed on the 19th day. Food intake, water intake and body weight of mice were recorded daily; the histopathological changes of jejunum were observed by HE staining; the expression of proliferating cell nuclear antigen (PCNA) in jejunum was observed by immunohistochemistry; the apoptosis of jejunum cell was detected by Tunel method; the contents of IL-6, IL-1β and TNF-α in jejunum tissue were determined by enzyme-linked immunoassay (ELISA).
    Results Compared with model group, high dose of M. oleiferia seeds water extract group (200 mg/kg) could alleviate the weight loss of mice significantly; extremely significantly increase the villous length of jejunum, decrease the crypt depth, and increase the ratio of villous length to crypt depth; increase the expression of PCNA in jejunum; reduce the degree of apoptosis; inhibite the expression of IL-6, IL-1β and TNF-α.
    Conclusion M. oleifonica seeds water extract can ameliorate 5-FU-induced jejunal mucositis, and with a dose-dependent manner.
