PurposeTo study the effect of leptin (LEP) on the mRNA expression of lipid droplet related genes FITM2, PLIN1 and PLIN2 in porcine primary adipocytes.
MethodsSubcutaneous primary adipocytes from DLY pigs at 15-day was treated with different mass concentrations LEP including 0, 50, 100 and 150 ng/mL for 48 hours, and the content of triglyceride (TAG) in cells were measured, and the mRNA expression levels of lipid droplet related genes FITM2, PLIN1 and PLIN2 were detected using qPCR technology.
ResultsCompared with 0 ng/mL LEP, the content of TAG and the mRNA expression levels of FITM2 and PLIN2 were significantly increased in 100 ng/mL LEP (P<0.05), the mRNA expression level of PLIN1 was significantly down-regulated (P<0.05). TAG content of 150 ng/mL LEP was significantly lower than 50 and 100 ng/mL LEP (P<0.05), and the mRNA expression levels of PLIN1 and PLIN2 were significantly higher than 50 and 100 ng/mL LEP (P<0.05).
ConclusionLEP can promote the mRNA expression of FITM2 and PLIN2, it can promote or inhibit the mRNA expression of PLIN1, however, there are differences in the mass concentration of LEP in promoting or inhibiting the expression of lipid droplet related genes.
