Optimization of the Microspore Culture System in Turnip (Brassica rapa var. rapa)

  • PurposeOptimizing the microspore culture system in turnip to improve the embryo yield, providing an important basis for the purification of turnip varieties and germplasm innovation.
    MethodAfter treatments at low temperature (4 ℃) and subsequent high temperature (33 ℃) for 24 hours respectively, one variety of turnip with a higher embryo emergence rate were screened from 54 varieties of turnip, and the microspore culture system of turnip was optimized by adjusting the ratio of activated carbon-agarose and the ratio of plant hormones.
    ResultsMicrospore embryogenesis of the variety KTRG-B-15 was the highest (25 embryos/30 buds). High mass concentrations of agarose could inhibit microspore embryogenesis. Microspore embryogenesis was ashigh as 137 embryos/30 buds after 0.1 mg/L NAA and 0.2 mg/L 6-BA were used in culture medium.
    ConclusionThe optimization of microspore culture system effectively improves the number of microspore embryogenesis in turnip, which is 5.48 times higher than that before optimization.
