PurposeTo study the effect of the combination of Dendrodium candidum extract and black tea extract on bone morphometric parameters in ovariectomized mice.
MethodsA mouse model of osteoporosis was established, and those mice were randomly divided into six groups, including a sham operation group (S group, unexcised ovary), a model group (M group), an estradiol group (E2 group, 1.5 mg/kg), the combination of D. candidum extract and black tea extract low dose group (DCE+BTE-L group, 0.475 g/kg), medium dose group (DCE+BTE-M group, 0.950 g/kg) and high dose group (DCE+BTE-H, 1.425 g/kg). After three months of continuous administration of the test substances, the left femur morphometric parameters of osteoporosis model mice was analyzed by Micro-CT.
Results The combination of D. candidum extract and black tea extract treated mice could improve bone trabecular structure, increase bone mineral density (BMD), bone volume/total volume (BV/TV) and bone trabecular thickness (Tb.Th), as well as decrease bone trabecular separation (Tb.Sp); among them, compared with the M group, the DCE+BTE-M group had the best effect, which obviously improved the trabecular bone structure, and BMD values increased by about two times (P<0.05), BV/TV increased by about one time.
ConclusionTreatment of osteoporosis model mice with the combination of D. candidum extract and black tea extract obviously improved bone microstructural degeneration induced by ovariectomy.
