Study on in vitro Cultural Technique of Red Pitaya

  • Purpose In order to provide technical support for large-scale seedling raising, the in vitro cultural technique was studied to establish the technical system of tissue culture and massive propagation of red pitaya.
    Method The young stem segments of red pitaya were used as explants, and Murashige and Skoog (MS) medium was supplemented with different ratios of exogenous hormones to select the suitable media component for induction of adventitious shoots, proliferation, strong seedling culture and rooting induction, respectively.
    Results The best disinfection method was to use 75% alcohol to disinfect for 10 seconds, rinse with sterile water once, disinfect with 0.1% mercury bichloride for 4.5 minutes, and rinse with sterile water 5-8 times. The optimum medium for the adventitious shoot induction was MS+6-BA 3.0 mg/L+NAA 1.5 mg/L+sucrose 30 g/L+agar 4.6 g/L. The germination rate was as high as 87.03%; the formula for the optimal adventitious shoot proliferation medium was MS+6-BA 3.0 mg/L+NAA 1.5 mg/L+active carbon powder 0.2 g/L+sucrose 30 g/L+agar 4.6 g/ L, the proliferative coefficient of adventitious shoots was as high as 7.33; the optimum culture medium for rooting induction was 1/2MS+NAA 0.5 mg/L+IBA 0.5 mg/L+carbon powder 0.2 g/L+sucrose 30 g/L+agar 4.6 g/L, the rooting rate was 100%; the suitable transplanting substrate was V (perlite)∶V (laterite)∶V (humus) = 1∶1∶1, and the survival rate of the plantlets was as high as 98.9%.
    Conclusion The optimum tissue culture system of red pitaya with stem segments as explants was established. Under this system, high rates of adventitious shoot induction, shoot proliferation, rooting and survival of plant transplantation were obtained.
