PurposeThe pollen viability was studied in order to provide the technical support for cross breeding of Pinus kesiya var. langbianensis.
MethodThe effects of sucrose, H3BO3 and CaCl2 single factor and orthogonal test on the pollen germination and pollen tube growth of P. kesiya var. langbianensis in vitro were studied.
ResultsThe pollen germination and pollen tube growth would be enhanced when the concentration of sucrose, H3BO3 and CaCl2 of the culture medium were within a certain range. The single suitable concentration of sucrose, H3BO3 and CaCl2 for pollen viability test were 10%, 100 mg/L and 100 mg/L, respectively. Sucrose had significant effect on the pollen germination and pollen tube growth in orthogonal test. The optimum culture medium which support the germination and tube growth was 10% sucrose +30 mg/L H3BO3+100 mg/L CaCl2.
ConclusionThe culture medium suitable for detection of pollen viability of P. kesiya var. langbianensis was obtained.
