Analysis of Microsatellite DNA Markers Emphasis on Correlation with Some Economically Important Traits in Mandarin Fish Hybrid Siniperca chuatsi×Siniperca scherzeri

  • PurposeThe aim was to analyze the genetic diversity and screen out the microsatellite markers related to growth traits of mandarin fish hybrid (Siniperca chuatsi×Siniperca scherzeri).
    MethodThe genetic diversities of 96 mandarin fish hybrid were analyzed based on 15 high polymorphism microsatellite markers by using the software of PopGene 3.2. The deviation of Hardy-Weinberg balance was analyzed using Chi-square test. The correlation between microsatellite markers and the mainly growth traits was analyzed using the least square method of the general linear model in SPSS 16.0.
    ResultTotal of 190 different alleles were observed, with alleles ranging from 3 to 22 per locus and the numbers of the effective alleles ranging from 1.417 7 to 10.326 1. The observed heterozygosity was found to be from 0.260 4 to 0.914 2, with mean value of 0.601 4. The expected heterozygosity was found to be from 0.296 2 to 0.914 2, with mean value of 0.742 3. The polymorphism information content ranged from 0.279 5 to 0.903 2, with mean value of 0.717 7. Only 2 loca were not significantly deviated Hardy-Weinberg balance (P>0.05). Correlation analysis results showed that significant (P<0.05) or extremely significant (P<0.01) relationships were found between three growth traits including body weight, body length and body height and five microsatellite markers CD90, MDJ821, PY21, PY55 and 72217.
    ConclusionThe results indicated that alleles had uniform distribution and high polymorphism in the whole population, which is suitable for the study of genetic diversity of mandarin fish hybrid. The DNA fragment length 219 bp of microsatellite markers 72217 can be molecular marker to assist breeding of the mandarin fish hybrid.
