PurposeTo optimize of enzymatic hydrolysis and purification conditions of 4-(α-L-rhamnosylbenzyl) isothiocyanate (GMG-ITC) from Moringa oleifera seeds.
MethodsThrough single factor and orthogonal experiments, we optimized the conditions for enzymatic extraction of glucosinolates from the de-oiled M. oleifera seeds. Then, the high purity GMG-ITC by extraction and recrystallization with methylene chloride were obtained.
ResultAn optimum condition for enzymatic hydrolysis was obtained as following: material to solvent ratio 1∶60 (g∶mL), temperature 30 ℃, reaction time 9 h, pH 5, by HPLC analyzing.
ConclusionWe described a simple and efficient four-step process including de-oiling, enzymatic hydrolysis, extraction and recrystallization to afford GMG-ITC from M. oleifera seeds, an overall yield of 4.70% was achieved with 98% purity.
