Genetic Diversity Analysis of Introduced Olive Germplasm Resources in Yunnan by SSR Markers

  • Purpose The confusion, homonyms and homonyms among different varieties of Olea europaea in Yunnan were accurately identified.
    Method Genetic diversity and molecular identification of 82 olive samples collected from Yunnan, Yongren and Lijiang were studied by SSR fluorescence labeling technique.
    Results 181 alleles were detected by 20 pairs of primers, 3–14 alleles were detected on each pair of primers, average of 9.05. Heterozygosity of the whole population ranged from 0.138 1 to 0.822 6, with an average of 0.489. The expected heterozygosity was between 0.509 8 and 0.871 5, with an average of 0.721 6, and 3 primers with PIC values above 0.8, they were DCA09, DCA18 and GAPU71B, respectively. The amplified product fragments of 20 loci are between 53 bp and 233 bp. According to Nei’s genetic distance among varieties, construct the dendrogram of all varieties by using UPGMA method, the results of 82 olive oil samples tested were clustered into 9 categories, breeding and varieties are not separate, but most have the same name of a variety of materials are in the same group.
    Conclusion Yunnan oil olive cultivars and varieties have high levels of genetic diversity.
