Study on the Micropropagation of Plumeria rubra var. acutifolia

  • Purpose The micropropagation of Plumeria rubra var. acutifolia was studied in order to provide the technical support for the rapid propagation of superior varieties (lines).
    Method The technology system of micropropagation was established on 1/2 MS medium plus cytokinin (6-BA) and auxin (NAA and IBA) using the terminal buds of sterile seedlings as explants.
    Results The suitable medium for induction of adventitious bud was 1/2 MS+1.0 mg/L 6-BA+0.1 mg/L NAA, and the proliferation rate of bud was 4.2-fold. The suitable medium for prolongation of plantlets was 1/2 MS+0.05 mg/L NAA. The suitable medium for induction of adventitious root was 1/2 MS+3.0 mg/L IBA, and the rooting rate of plantlets was 93.3%. The mixed substratum with perlite and peat soil (1:2, volume ratio) was good for transplanting in acclimatization stage, and the survival rate of transplanted plantlets was 86%.
    Conclusion The optimum medium and suitable transplanting medium for adventitious bud induction, elongation and rooting were obtained, and the micropropagation system was established in P. rubra var. acutifolia.
