[Purpose] The aim of this study was to determine the associations between the mutations in leukemia inhibitory factor (LIF) gene and the reproductive traits of sows.[Method] The polymorphisms of C4950G and C6988T within LIF gene were examined by PCR product directed sequencing in 405 Large White pigs. Combining the data of 1716 litters reproductive performance, the effects of genotypes for single locus and combined genotypes for bi-loci on 5 reproductive traits were analyzed.[Result] The allele with the highest frequency at locus C4950G and C6988T was G and T, respectively, both loci were in Hardy-Weinberg equilibrium and had a higher heterozygosity. At the C4950G locus, sows of GG genotype had higher total number born (TNB), number of piglets born alive (NBA), litter weight at birth (LWB) and number of alive piglets on 21st day (N21) when compared to the other two genotype (P<0.05 or P<0.01); and at the C6988T locus, the above traits of sows with CT genotype were significantly higher than those of TT genotype (P<0.05). The obvious synergistic action was showed on the effects of the two loci on the sow reproduction traits. Compared to other combined genotypes, TNB, NBA and N21 of the sows with genotype GGCT increased 0.90-1.05, 0.54-0.88 and 1.02-1.95 piglets, respectively (P<0.05 or P<0.01), together with larger LWB and litter weight of piglets on 21st day (P<0.05 or P<0.01).[Conclusion] The present results further confirmed the significant associations of LIF gene with sow reproductive traits, but its specific effects would be variety in different pig breeds or populations.
