云南省粘虫的遗传多样性

摘要 粘虫(Mythimna separata)是重要的季节性迁飞的农业害虫,为弄清云南省粘虫的遗传多样性,本研究利用ISSR分子标记技术分析了云南省23个地理种群粘虫的遗传多样性和遗传结构特征。结果表明:100条ISSR引物中共筛选出9条多态性引物,这些引物在23个地理种群粘虫中扩增出198个位点,各种群多态性位点百分比(P)变化范围为44.95%~66.16%,其中以西畴县西洒镇(XS)种群的多样性位点最高,弥渡(MD)种群最低;种群遗传分化系数(Gst)为0.3,表明30%的遗传变异发生在种群间,70%的变异来源于相同种群不同个体之间;基因流(Nm)为1.669、平均遗传相似系数为0.898 2,UPGMA聚类分析结果表明23个地理种群遗传距离与地理距离相关性不显著(r=-0.085 6,P=0.282 0)。综合遗传多样性结果,云南省粘虫的遗传多样性较高,各地理种群间产生了一定程度的遗传分化;各种群遗传结构相似性较高,居群间变异较小,居群内变异较大;不存在由遗传漂变引起的种群间遗传分化,该研究结果可为云南省粘虫防控策略的制定提供新的思路和方法。

Abstract: The oriental armyworm, Mythimna separata (Walker) is a major agricultural and migratory pest in Yunnan Province. To verify the genetic diversity of oriental armyworm, the genetic diversity and genetic structure of 23 M. separata populations in Yunnan Province were analyzed by inter-simple sequence repeat (ISSR). The results showed that 9 primers were selected among 100 primers and 198 loci were detected by ISSR marker analysis. The percentage of polymorphic loci (P) was 44.95%-66.16%, the Xisa population was the highest among 23 populations, the lowest polymorphic loci was in the Midu population. Coefficient of gene differentiation (Gst) among 23 populations was 0.3, showed that the genetic variation among individuals in the same population (70%) was higher than that of among populations (30%). Gene flow (Nm) among those populations was 1.669, the average of genetic similarity was 0.898 2, and the Mantel test results suggested that there was no significant correlation between the genetic distance and geographic distance of different M. separata populations(r=-0.085 6,P=0.282 0). Overall, the genetic diversity of armyworm in Yunnan Province was high, and there was genetic differentiation among different geographic populations. Moreover, the population genetic structure similarity was high and armyworm could prevent genetic drift caused by genetic differentiation among those 23 populations. The results in this paper can provide some new ideas for integrated pest management of M. separata.
